A high magnification image of a nemaline rod closely associated with an adjacent Z-line showing its substructure and osmiophilia. Nemaline rods exhibit a highly organized crystalline lattice-like structure. In longitudinal sections, they show axial periodicity (roughly 8–10 nm) and striations both parallel and perpendicular to the long axis. (electron micrographs) ---- Diagnosis: Nemaline rod myopathy. Rods are evident on routine histochemistry, toluidine blue stained plastic sections and on ultrastructural analysis. A phalloidin stain is positive for the presence of actin in the nemaline rods..
