1C1-3 The NADH (nicotinamide adenine dinucleotide) stain in skeletal muscle highlights mitochondria and oxidative enzyme activity. In this histochemical stain, Type 1 (slow twitch) fibers stain very dark as they are loaded with mitochondria resulting in a highly oxidative environment, while Type 2 (fast twitch) fibers stain much lighter/paler reflecting their reliance primarily on glycolysis for quick energy bursts. Target fibers have a lucent center and a dark rim of increased activity. (NADH)
