Ki-67 proliferation index is elevated, highlighting 30-40% of the
tumor cells. ---- Ancillary workup (not shown): Neurofilament immunohistochemistry is negative in the tumor indicating a solid growth pattern. Synaptophysin shows focal immunoreactivity. Mutant BRAF V600E immunohistochemistry is negative. Alk-1 IHC is negative. ---- FISH studies were negative for ALK, ROS1, and BRAF gene rearrangements and MET gene amplification. Sendout FISH studies were negative for FOXR2, BCOR, NTRK1, NTRK2, and NTRK3 gene rearrangements. However, targeted molecular sequencing by Foundation One CDx identified an ETV6-NTRK3 fusion. T---- umor methylation profiling was performed at the NIH/NCI laboratory and revealed a consensus match to Infant-type hemispheric glioma using the Heidelberg and NCI classifiers. ---- The final diagnosis in this case was: Infant-type hemispheric glioma, NTRK-altered.
